Mechano growth factor (MGF)

MGF, IGF-1Ec, PEG-MGF

Stretch a muscle hard and it starts reading its IGF-1 gene differently. MGF is the name given to that loading signal, and the synthetic peptide sold under the same name has a far shakier record than the biology behind it.

Mechano growth factor (MGF), in short

Mechano growth factor (MGF) is a version of the IGF-1 gene product that muscle switches on after stretch or heavy loading. A synthetic 24-amino-acid piece of it has been tested on muscle stem cells in the lab, with conflicting results, and a pegylated form, PEG-MGF, is sold online. None of the sources here describes a human trial of injected MGF.

How does Mechano growth factor (MGF) work?

Muscle makes MGF after hard loading. In cell tests, it acts a little differently than IGF-1.

  • Makes repair cells multiply

In a cell test, its active piece made muscle stem cells multiply and kept them from maturing too soon.1

  • Scientists disagree

The first study found it did not need the usual growth signal. A later lab disagreed, and a replication found no effect.1810

  • Young cells last longer

In human muscle stem cells, it let cells from young muscle keep dividing longer, but not cells from old muscle.3

Mechano growth factor (MGF) benefits: what it is studied for

All of this is from cells, mice, or measurements of the body’s own MGF. No source here describes a trial of injected MGF in people.

Where MGF came from: a stretched rabbit muscle

In the mid-1990s, Geoffrey Goldspink’s lab in London did something simple to rabbit muscle. They held it under active stretch, which makes muscle grow fast, and then looked at which versions of the IGF-1 gene it was reading. Resting muscle made one familiar form. Stretched muscle added a second form that was barely detectable without the extra load.

The difference lay in the tail. The new variant carried a 52-base-pair insert in the stretch of genetic code for the E domain, the end piece of the IGF-1 precursor protein. That insert shifts the reading frame, so the tail of the resulting protein is different. In rabbits and rodents it is called IGF-1Eb. The human equivalent, with a 49-base-pair insert, is IGF-1Ec.

Because it appeared in response to mechanical load, the team named it mechano growth factor. The same 1996 paper detected the human form in muscle after changes in physical activity lasting from two hours to six days, while very little appeared in muscle that had no extra stretch or activity.

That part of the biology is well supported. Muscle does change how it splices the IGF-1 gene when it is loaded. The harder step is the leap from a gene variant to a synthetic peptide in a vial, and to see why, you have to follow what researchers made and what they found.63

02 / 10

How mechano growth factor is thought to work

The IGF-1 gene makes a precursor protein. The front portion becomes mature IGF-1, the 70-amino-acid hormone, and the tail, called the E peptide, can be cut away. Different splice variants give different tails. The synthetic peptide sold as MGF corresponds to the last 24 amino acids of the Eb tail, the version that matches human Ec.

In 2002, Yang and Goldspink reported that this E domain did something mature IGF-1 does not. In a muscle cell model, it increased myoblast proliferation while holding back terminal differentiation, the step where young muscle cells fuse into fibers. Blocking the IGF-1 receptor with an antibody did not stop the effect, so they proposed that MGF works through a separate receptor.

The model was elegant. After injury or heavy loading, an early burst of MGF would expand the pool of satellite cells, the muscle’s resident stem cells, and a later rise in the standard IGF-1Ea form would push those cells to mature and fuse. A 2010 review describes that timing in injured muscle, with IGF-1Eb up first and IGF-1Ea later.

In 2011, a Paris team tested the 24-amino-acid peptide on human muscle stem cells from donors of different ages. It extended the cells’ dividing life span and delayed senescence in cells from newborn and young adult muscle, though not from old adult muscle. The authors suggested it might help against age-related muscle loss without the cancer-promoting effects seen with IGF-1.10713

MGF and exercise: what happens in human muscle

The strongest human evidence concerns the body’s own MGF, measured in muscle biopsies. In a 2003 London study, eight young adults aged 25 to 36 and seven older adults aged 70 to 82 did ten sets of six heavy single-leg knee extensions. Biopsies taken two and a half hours later compared the worked leg with the rested one.

At rest, MGF messenger RNA was about 100 times lower than the standard IGF-1Ea form, and resting levels were similar across ages. After the exercise, MGF rose in the young group but not in the older one, while IGF-1Ea did not change in either. The authors read this as an age-related loss of sensitivity to mechanical loading.

A Finnish study in 2016 complicated the picture. Seventeen men, seven younger and ten older, did 21 weeks of resistance training, with biopsies taken 48 hours after test sessions before and after the program. MGF levels and the related signaling proteins did not differ between age groups and did not change systematically with training.

The two studies used different exercises, different ages and very different biopsy timing, two and a half hours versus two days, so they may have caught different moments. Both are small. Together they show that loading changes IGF-1 splicing in human muscle, and they leave open whether aging blunts that response.24

The MGF peptide nobody had found in the body

The synthetic MGF peptide, those 24 amino acids, had not been shown to exist on its own in a living body when a 2010 review in Endocrinology took stock. The review stated that no analogous peptide product of the IGF-1 gene had been identified in or isolated from cultured cells, the fluid they grow in, animal tissues or body fluids. The review’s title calls MGF a putative product of IGF-1 gene expression.

What muscle does contain is revealing. When a University of Pennsylvania team measured the forms of IGF-1 in mouse muscle in 2013, the predominant forms were pro-IGF-1 with the E peptide still attached, some of it carrying sugar groups, rather than mature IGF-1. In the mouse muscle examined, the tail mostly stayed on the parent molecule.

That matters because the MGF story assumes the tail is cut loose and travels as its own signal. If it mostly stays attached, the gene variant may change how pro-IGF-1 behaves, and a free-floating 24-amino-acid peptide may be largely a laboratory construct. The 2013 study did test part of this directly: pro-IGF-1 without sugar groups activated the IGF-1 receptor about as well as mature IGF-1, while the sugar-coated form activated it much less. Whether free 24-amino-acid MGF plays a natural signaling role is a separate question, and still open.

The 2010 review asked readers to separate what synthetic MGF does in a dish from anything the IGF-1 gene is known to make. Anyone weighing a vial labeled MGF should make the same separation. The loading signal in your muscle is real. Whether the peptide in the vial is a faithful copy of it is an open question.79

Does MGF work? The replication problem

Lab findings earn trust when other labs can repeat them. In 2014, scientists at two drug companies, interested in MGF as a possible route to better muscle regeneration, set out to reproduce its claimed effects on mouse and human myoblasts. They tested the native peptide across a range of concentrations and a stabilized version as well.

MGF failed to increase proliferation of C2C12 mouse muscle cells or primary human myoblasts. It failed to delay their fusion into myotubes. It had no significant effect on primary mouse muscle stem cells, and no activating effect in heart muscle cells. Mature IGF-1 and full-length IGF-1Eb made the myoblasts proliferate, and IGF-1 drove a strong response in the heart muscle cells. The authors wrote that the results call into question whether MGF has a physiological role.

A separate Pennsylvania lab found effects of a different kind. Synthetic E peptides did increase signaling, myoblast proliferation and migration, yet blocking the IGF-1 receptor shut those effects down. The E peptides did not activate the receptor themselves. They made more of it available on the cell surface, so IGF-1 worked better. The authors proposed that E peptides have little independent activity.

So the core claim is split three ways. One group reports independent stem cell effects through a separate receptor, a second finds no effect, and a third finds effects that run through the ordinary IGF-1 receptor. When independent labs disagree this sharply about cells in a dish, confident claims about injections in people run well ahead of the science.1081

Bigger muscles, weaker muscles: the mouse result

The most sobering MGF-related experiment did not use the synthetic peptide at all. It used gene delivery. In 2014, the Pennsylvania group used viral vectors to make mouse muscle produce E peptides continuously, with the mature IGF-1 portion deliberately disabled so only the tail could act. The EB peptide, the rodent counterpart of human MGF, increased satellite cell proliferation and produced significant muscle growth.

The muscles got bigger and weaker. The extra mass came with a loss of strength, and the growth depended on the IGF-1 receptor, the same pathway mature IGF-1 uses. The authors concluded that sustained EB expression may drive hypertrophy but has significant physiological consequences for muscle.

For anyone training, that gap between size and function is the whole point. Muscle that grows without getting stronger has added tissue that does not do the job muscle exists for. This is one mouse study using gene delivery, so it cannot be mapped straight onto a person injecting a peptide. It is still one of the few tests of sustained E-peptide signaling in a living animal, and its answer was mixed.113

MGF side effects and the PEG-MGF problem

A 2026 clinical review lists PEG-MGF among the peptides met in clinics and online self-administration protocols. It is the peptide with polyethylene glycol attached, a common way to make a peptide last longer in the body. FDA reviewed it as a nominated compounding substance, and its safety page, current as of April 22, 2026, lists PEG-MGF among substances that may present significant safety risks.

FDA’s concerns are specific. It says compounded PEG-MGF may carry a significant risk of immunogenicity, meaning the immune system may react against it, and may contain impurities that are hard to characterize. It also states that it has not identified any human exposure data and lacks important information on whether the substance would cause harm to people.

Because MGF comes from the IGF-1 gene, the cautions that apply to IGF-1 are a sensible starting point. Higher circulating IGF-1 is linked in large analyses to a modestly higher risk of prostate and premenopausal breast cancer, and IGF-1 itself lowers blood sugar. The Paris researchers hoped the E peptide might avoid IGF-1’s cancer-promoting effects. That hope is untested in people, and the mouse data showing growth through the IGF-1 receptor give reason for caution.

A 2026 clinical review of performance peptides places PEG-MGF among compounds with no regulatory approval and uncertain product composition. Across the growth hormone and IGF-1 peptides it covers, it lists reported problems including blood sugar disturbances, fluid retention, muscle and joint aches and injection-site reactions. With no human data on MGF itself, its own side effect profile is unknown.51412311

Which labs matter if you are curious about MGF

IGF-1 itself has well-known effects on fuel handling. Whether synthetic MGF or PEG-MGF shares those effects in people is unknown, so metabolic markers are simply a sensible general baseline. Fasting glucose is your blood sugar after a night without food. Fasting insulin shows how hard your pancreas works to hold that level, and it can rise before glucose does. HbA1c is your average blood sugar over roughly three months.

Glucose and HbA1c assess blood sugar directly, while fasting insulin needs context to interpret. None of this is a validated way to judge MGF safety. It is useful background for anyone training hard, since muscle is the major site of insulin-stimulated glucose disposal and the IGF-1 system sits right at that intersection of growth and fuel.

Higher circulating IGF-1 is linked to prostate cancer risk in large analyses, but that does not make PSA a way to check MGF safety. PSA can rise for many noncancerous reasons, and whether to test is a screening decision to make with a clinician. Vitals Vault starts peptide care with a blood panel at Quest Diagnostics reviewed by a licensed clinician, and for MGF the useful step is understanding your own markers before considering anything else.12

Is MGF legal? Where it stands in 2026

MGF has never been approved by FDA for any use. PEG-MGF appears on FDA’s list of compounding substances that may present significant safety risks, and it was not among the six peptides that FDA allowed 503A pharmacies to compound under enforcement discretion on October 29, 2026. There is no prescription pathway, and vials sold online as research chemicals are not made or tested for people.

In sport, the World Anti-Doping Agency’s 2026 Prohibited List names mechano growth factors under S2, growth factors and growth factor modulators, banned in and out of competition.141315

What this means for you

Mechano growth factor began as a genuine discovery. Loaded muscle reads its IGF-1 gene differently, and in young adults the MGF form rises within hours of a heavy session. The synthetic peptide built from that discovery has a far weaker record. As of a 2010 review, it had not been found on its own in the body, independent labs disagree about whether it does anything in cells, and its pegylated form has no human exposure data at all.

Heavy resistance exercise can raise MGF mRNA under some conditions, and training backed by enough protein and sleep builds muscle whether or not MGF is the reason. That route comes with the strength gains that the mouse gene study did not get.

So the useful question is how well your muscle is responding to the loading you already give it. Are you progressing in strength, recovering between sessions, and keeping your glucose and insulin markers where a clinician would want them?27101411

What to expect with Mechano growth factor (MGF)

There is no supervised path to MGF or PEG-MGF, so the realistic process is a different one. A clinician-led route starts with a history and baseline labs, then focuses on training, recovery and approved options for whatever goal is driving the interest.

On timing, the human data concern the body’s own MGF. In young adults it rose within two and a half hours of a heavy session, and the stimulus is free and repeatable. Studies have not established a timeline for any effect of injected MGF in people.214

Mechano growth factor (MGF) myths and evidence

  • Injecting MGF just tops up the growth factor your muscles already make

Loaded muscle makes more of the MGF gene variant, but a 2010 review found no free MGF peptide identified in tissues or body fluids. In mouse muscle, pro-IGF-1 forms with the E peptide still attached predominated.79

  • MGF is proven to activate muscle stem cells

Some cell studies found that it did. Scientists at two drug companies could not reproduce the effect in mouse or human myoblasts or in mouse muscle stem cells, and another lab found the effects depended on the IGF-1 receptor.3108

  • More muscle from MGF means more strength

In mice given gene constructs that kept making rodent E peptides, with mature IGF-1 disabled, muscles grew but lost strength. That was gene delivery, not a test of injected synthetic MGF.11

  • MGF is safer than IGF-1 because it does not cause cancer

That is a hypothesis from cell work. MGF has no human safety data, FDA flags immunogenicity and impurity risks for PEG-MGF, and the mouse growth effect ran through the IGF-1 receptor.31411

How is Mechano growth factor (MGF) taken?

MGF and PEG-MGF sold outside medicine are injected. There is no approved product and no prescribing pathway, so no clinician can legitimately provide it.145

Mechano growth factor (MGF) dosage

There is no typical human dose of MGF or PEG-MGF. No FDA label, human trial or clinical review gives one, and FDA notes it found no human exposure data for PEG-MGF. Online dosing charts are not based on studies in people. If a clinician ever discusses it, your clinician sets your dose.14

Typical Mechano growth factor (MGF) dosage

No human dose of MGF or PEG-MGF has been published. The human research only measures the body’s own MGF in muscle after exercise, and FDA’s compounding safety page says it identified no human exposure data for PEG-MGF. Your clinician sets the dose of any treatment you take.

These are reference doses from FDA labels, studies in people and clinical practice reports, shown for education. They are not a recommendation or a prescription. Your dose, form and schedule should be set by a licensed clinician who knows your history and lab results.

Muscle talks to itself before the rest of the body listens

Resistance exercise does not only raise circulating hormones. Loaded muscle also changes how it reads its own IGF-1 gene. MGF is part of that local signaling and is made where the stress is.

That makes the case for injecting it unclear. A signal made locally in a stressed muscle is not the same as a dose given from outside. The studied way to raise natural MGF is to load the muscle, and even that response varied between studies.2164

Mechano growth factor (MGF) questions

What is mechano growth factor (MGF)?

MGF is a splice variant of the IGF-1 gene, also called IGF-1Ec, that skeletal muscle produces after mechanical stress. A synthetic E-domain peptide and a pegylated form (PEG-MGF) are sold online.

What is MGF used for?

In research it is studied for activating muscle stem cells and for muscle repair. People who seek it out online usually want muscle growth and recovery. It is not FDA-approved for any use.

What are the benefits of MGF?

In cultured muscle cells the E peptide increased stem cell proliferation, and natural MGF rises after heavy training. No benefit from injected MGF has been shown in any human trial cited here.

How does MGF work?

In early cell studies, the E domain increased myoblast proliferation and delayed differentiation, apparently through a receptor other than the IGF-1 receptor. Later work disputes this. One lab found the effects ran through the IGF-1 receptor, and scientists at two drug companies found no effect on muscle cells.

What are the side effects of MGF?

Human safety data do not exist. FDA flags immunogenicity and impurity concerns for PEG-MGF, and general IGF-1 cautions on blood sugar and cancer risk are sensible until data show otherwise.

What is the MGF dosage?

There is no typical human dose, because injected MGF and PEG-MGF have never been tested in a human trial. Online charts are not based on studies in people. Do not use any chart to dose yourself; your clinician sets your dose.

MGF vs IGF-1, what is the difference?

Both come from the IGF-1 gene. MGF is a splice variant with a distinct E domain made locally in muscle after load. Mature IGF-1 is circulating and acts through the IGF-1 receptor.

What is PEG-MGF?

PEG-MGF is MGF modified with polyethylene glycol to last longer. FDA lists it on its compounding safety page, noting immunogenicity risk and no identified human exposure data.

Is MGF FDA approved or legal?

It is not FDA-approved for any use. It was not among the six peptides covered by FDA’s October 2026 compounding policy. Products sold online as research chemicals are not made or tested for people.

Is MGF banned in sports?

Yes. WADA’s 2026 Prohibited List names mechano growth factors in S2, prohibited in and out of competition.

Can I get MGF through Vitals Vault?

No. MGF is not an approved or compounding-eligible medicine. Vitals Vault is opening clinician-led peptide care that starts with a blood test, and you can join the peptides waitlist to hear first.
