LH and testosterone in men
Kisspeptin-10 given to healthy men raised LH in a dose-dependent way, and a 22.5-hour infusion raised testosterone from 16.6 to 24.0 nmol/L.1

Kisspeptin was first found as a cancer gene in Hershey, Pennsylvania. Seven years later it turned out to be the switch that starts puberty, and kisspeptin-10 is the short fragment researchers use to flip it.

Kisspeptin-10 is the shortest fully active piece of kisspeptin, the brain hormone that switches on GnRH neurons and so sets off LH, FSH and sex hormone release. In small human studies it raised LH and testosterone in healthy men and in men with type 2 diabetes and low testosterone. It is studied for fertility and reproductive hormone disorders, with early human evidence.

It works high up in the brain, where the signal that starts the reproductive hormones begins.
It switches on the brain cells that release the hormone that starts the reproductive chain.78
In healthy men, it raised a pituitary hormone within minutes, and a longer infusion also raised testosterone.1
Constant exposure blunts the response. On-and-off exposure keeps it, like the body’s own pulses.24
The evidence is early but human. Studies measured hormone responses in small groups, and none tests kisspeptin-10 for pregnancy or symptom relief. The IVF card used the longer form, kisspeptin-54.
This guide cites no findings at this level.
Shows a mechanism
Mice without the kisspeptin receptor also fail to go through puberty. This shows the system matters, but it is not a test of kisspeptin-10.56
Shows an effect in a body
Kisspeptin-10 raised hormones in small groups of healthy men, including for 12 days with on-and-off infusions. No trial shows pregnancy or symptom relief from it.12
Shows it works in people
Kisspeptin has real human reproductive-endocrinology research behind it, with most studies using infusions in small groups of healthy men or in women with hypothalamic amenorrhea. A 2026 review of kisspeptin approaches for testosterone-therapy suppression found direct evidence for restoring fertility remains limited.1249
The full guide to Kisspeptin-10
In 1996, cancer researchers at Penn State’s College of Medicine in Hershey, Pennsylvania, a town better known for its chocolate kisses, were hunting for genes that stop melanoma from spreading. They found one, named it KiSS-1, and showed that putting it back into melanoma cells suppressed their ability to metastasize. In healthy tissue, its highest expression was in the placenta, an odd address for a cancer gene.
For a few years, nobody knew what its protein did in a healthy body. In 2001, a team in England cloned a human brain receptor closely related to a rat orphan receptor called GPR54 and found that peptides made from KiSS-1 activated it potently. The receptor’s gene was most highly expressed in the brain, the pituitary and the placenta.
Then, in 2003, two groups reached the same answer from opposite directions. In France, researchers studying a large family in which five siblings never went through puberty found that all five carried a deletion in the GPR54 gene. In Boston, a second team found GPR54 mutations in other patients with absent puberty and bred mice lacking the receptor, which had small testes or immature ovaries.
The finding reshaped reproductive endocrinology. Kisspeptin and its receptor, now called KISS1R, are essential for normal GnRH release and for puberty itself. A peptide first studied for stopping cancer spread turned out to sit at the top of the reproductive hormone chain.1011657
Reproduction runs on a relay. The hypothalamus releases GnRH in pulses, the pituitary answers with LH and FSH, and those two hormones tell the testes or ovaries to make testosterone or estradiol and to mature sperm or eggs. Kisspeptin sits one step above GnRH. Kisspeptin neurons in the hypothalamus, many of which also make a partner signal called neurokinin B, set the pace of those GnRH pulses.
The 2003 mouse work showed how specific that job is. Mice without the receptor still had normal amounts of GnRH stored in the hypothalamus, and they still responded when GnRH or gonadotropins were given. The machinery downstream, pituitary and gonads, worked. What was missing was the signal to start it.
Kisspeptin comes in several lengths. Kisspeptin-54 is the major form circulating in human blood, and kisspeptin-10 is the minimal sequence that keeps full activity at the receptor. In men, kisspeptin-54 has a half-life in blood of about 28 minutes, and human studies have used both single injections and infusions.
Reviews also describe kisspeptin acting on sexual behavior and attraction, the placenta and bone. Its neighbor, neurokinin B, drives menopausal hot flashes, and a drug that blocks its receptor, fezolinetant, is now approved for them. That approval shows how ready this small corner of the brain has become for medicine.8751413
The first study of kisspeptin-10 in people came from Edinburgh in 2011. Given as a single injection into a vein, it caused a rapid, dose-dependent rise in LH in healthy men, roughly tripling it within 30 minutes at the most effective dose. A higher dose produced a smaller response, an early hint that more is not always better with this signal.
Longer infusions showed what happens downstream. Over 22.5 hours, LH rose about fourfold and testosterone climbed from 16.6 to 24.0 nmol/L. A gentler infusion revealed the mechanism more clearly, raising LH pulse frequency from 0.7 to 1.0 pulses an hour and making each burst larger. That is consistent with kisspeptin-10 speeding up the body’s own GnRH rhythm, which cannot be measured directly in people.
A London group had already shown the same direction with the longer kisspeptin-54. In a 2005 double-blind crossover study of 6 men, a 90-minute infusion raised LH, FSH and testosterone compared with saline.
The practical question is whether the effect lasts. A 2026 Imperial College study of 15 healthy men, split across three experiments, gave kisspeptin-10 under the skin. In four men, five days of continuous infusion left LH and FSH no higher than placebo, although testosterone stayed raised. In seven others, intermittent infusions, running for part of each day, kept gonadotropins up across 12 days, and the receptor still responded at the end.132
Low testosterone is common in men with type 2 diabetes and severe obesity, and it is tempting to assume the testes have simply worn out. In 2010, the Edinburgh team proposed another explanation. Evidence pointed to the hypothalamus releasing less GnRH in these men, and they hypothesized that falling kisspeptin is the common pathway linking high blood sugar, inflammation, leptin and estrogen feedback to low testosterone.
In 2013 they tested the idea in a proof-of-concept study. Five men with type 2 diabetes and low testosterone, with an average BMI of about 40, were given kisspeptin-10. Their LH rose about as much as it did in seven healthy men. During an 11-hour infusion in four of them, LH pulse frequency increased and testosterone rose from 8.5 to 11.4 nmol/L.
In these five men, the pituitary and testes could still answer when called. That is consistent with a quiet signal upstream, though it does not prove missing kisspeptin is the cause. Picture a factory with a working production line and a phone that has stopped ringing. One way to read it is that some metabolic low testosterone is a message problem, and it explains why weight, blood sugar and inflammation keep turning up in a hormone story.
This is still a hypothesis built on very small studies, and it does not make kisspeptin a therapy for diabetes-related low testosterone. It does suggest two practical habits. Measure LH alongside testosterone, and count metabolic health as part of the hormone conversation instead of a separate topic.1213
Hypothalamic amenorrhea is the loss of periods because the brain stops sending its signal, while the ovaries remain healthy. In 2010, a London team gave kisspeptin-54 to women with the condition. Frequent injections lost their effect quickly, with the FSH response nearly gone by the second day.
Spacing made the difference. A less frequent schedule caused only partial desensitization, and after 8 weeks those women had significantly higher reproductive hormones than women given saline. No adverse effects were seen. The lesson runs through all kisspeptin work. The axis listens to rhythm, and constant exposure tunes it out.
The most clinical use so far is in IVF. In 2014, 53 women undergoing IVF received a single injection of kisspeptin-54 to trigger the final maturation of their eggs. Embryos were transferred in 92 percent, and 23 percent had a clinical pregnancy. Because kisspeptin works through the body’s own GnRH, reviewers describe it as a potentially safer, more physiological trigger for this step.
These studies used kisspeptin-54, the longer form. Their results are a guide to what the kisspeptin system can do, and they should not be assumed to transfer automatically to kisspeptin-10.4148
Kisspeptin’s reach goes beyond hormone levels. In a 2022 trial at Imperial College London, 32 premenopausal women with hypoactive sexual desire disorder received a 75-minute infusion of kisspeptin-54 on one day and placebo on another, while their brains were scanned as they watched erotic videos and looked at faces. Kisspeptin shifted activity in regions tied to sexual and attraction processing.
Some of those changes tracked with how the women felt. Kisspeptin’s boost to hippocampal activity during erotic videos was larger in women with more baseline distress about their sexual function, and its effect in a region called the posterior cingulate cortex correlated with lower sexual aversion. It was well tolerated, with no reported adverse effects.
A matching 2023 trial in 32 men with low desire found kisspeptin modulated the brain’s sexual-processing network, increased penile tumescence during sexual videos by up to 56 percent more than placebo, and raised happiness about sex. The authors suggested kisspeptin could become the first drug for low desire in men.
These were single infusions in a scanner, and brain signals are a long way from months of daily life. They do suggest that the hormone at the top of the reproductive chain also speaks to the parts of the brain that decide whether sex feels appealing.1516
Lay the human record side by side and it is a set of careful, small physiology studies. Most enrolled between 5 and 53 people, ran for hours to weeks, and measured hormones or brain signals. Together they show that kisspeptin can drive the human reproductive axis on demand. They do not yet show better fertility, better sex lives or symptom relief from kisspeptin-10 itself.
The question many men bring to kisspeptin is whether it can protect fertility on testosterone therapy, which suppresses LH, FSH and sperm production. A 2026 narrative review of peptide strategies for that problem found that kisspeptin approaches may support the body’s own hormone signaling, but that direct evidence of restored sperm production or fertility after testosterone suppression remains limited.
Much of the human work also comes from a small number of research groups, mainly in London and Edinburgh. That gives the field consistent methods, and it is also a reason to want independent replication before any of these findings are taken as settled.1131429
In short human studies, kisspeptin has been well tolerated. The 8-week kisspeptin-54 study in women observed no adverse effects, and the brain imaging trial in women reported none. The 2026 kisspeptin-10 study in men ran for 12 days. None of these tells us what happens over months or years.
The main known limit is tolerance. Constant or too-frequent exposure blunts the response. Frequent kisspeptin-54 injections produced complete tolerance in women, and five days of continuous kisspeptin-10 left gonadotropins no higher than placebo in men. A poorly designed schedule can leave it doing nothing for LH and FSH.
Product quality is the other issue. FDA has said compounded kisspeptin-10 may pose a risk of immunogenicity, meaning the immune system could react to it, and may contain peptide-related impurities that are hard to characterize. It also notes no, or only limited, safety information for the proposed routes.
Because kisspeptin raises sex hormones, people with hormone-sensitive cancers, anyone pregnant or trying to conceive, and anyone already on testosterone or another hormonal medicine should talk with a clinician first.415218
Kisspeptin research is really a study of a relay, and four blood tests show which runner has slowed. LH is the pituitary’s message to the gonads. In men, a low or normal LH alongside low testosterone points up the chain, toward the hypothalamus or pituitary. A high LH with low testosterone points down, toward the testes.
FSH is LH’s partner, driving sperm production in men and follicle growth in women. Total testosterone is the main output of the axis in men. Estradiol tracks the ovarian cycle in women and, in men, estrogen made mostly from androgens, which also feeds back on the brain.
Measured together and repeated, these four turn a vague complaint into a map. That is how Vitals Vault approaches hormone questions. A blood panel at Quest Diagnostics comes first, a licensed clinician reads it alongside your history, and a retest shows which way the numbers are moving.813
Kisspeptin-10 is not FDA-approved for any use. On September 29, 2023, FDA placed it in category 2 of bulk substances nominated for 503A compounding, citing immunogenicity risk, impurity concerns and limited safety information. It was not among the six peptides covered by FDA’s October 29, 2026 enforcement discretion, so that policy does not open a compounding route for it.
Sport has its own rule. The World Anti-Doping Agency’s 2026 Prohibited List names kisspeptin and its agonists under S2.2.1, testosterone-stimulating peptides prohibited in males, in and out of competition.181719
Kisspeptin is the signal at the very top of the reproductive chain, found in a hunt for melanoma genes and confirmed by families whose puberty never started. Kisspeptin-10, its shortest active piece, reliably raises LH and testosterone in men in small studies, and related work with the longer form points to roles in fertility treatment and in how the brain handles desire.
If low testosterone or irregular cycles are your question, the useful first step is to find out where the chain has gone quiet. LH and FSH read next to properly timed testosterone or estradiol, and your history, help a clinician tell whether the message or the gland looks like the problem, sometimes with more testing to settle it.518
In studies, the hormone response came fast. LH rose within about 30 minutes of an intravenous dose of kisspeptin-10, and testosterone rose over hours of infusion. Timing then decided everything, since continuous exposure faded within days while intermittent exposure held up over 12 days.
There is no approved product, so any clinical use would sit within research or close specialist care. The sensible frame is a measured one, with baseline LH, FSH and testosterone or estradiol first and repeat testing to see whether the chain is responding.12

It works in the brain. Kisspeptin acts on KISS1R to trigger GnRH release, which then prompts LH and FSH from the pituitary. That chain, not a direct hit on the testes, is the main pathway the research describes.78
They share the active end but differ in length. Kisspeptin-54 is the main circulating form in people, and most fertility and desire trials used it. Kisspeptin-10 is the minimal fully active fragment, studied mostly in men.141
Not yet. Trials in 2022 and 2023 found single infusions of kisspeptin-54 changed brain responses to sexual cues in people with low desire. Neither trial showed lasting improvements in sex lives.1516
There is no FDA-approved dose of kisspeptin-10 for any use. Human studies gave it into a vein, or under the skin by pump, in research units: single weight-based injections or infusions lasting hours. A continuous five-day infusion under the skin lost its effect on LH and FSH, though testosterone stayed raised, while eight-hour daily infusions kept LH up for 12 days. These were research protocols, not a self-administered regimen. Your clinician sets any dose.1132
No kisspeptin-10 dose is approved. Studies in men gave single weight-based injections into a vein, or infusions into a vein or under the skin lasting hours, in research units with frequent blood sampling, and a continuous five-day infusion lost its effect on LH and FSH while testosterone stayed raised. These are research protocols, not a home regimen, and a clinician decides any use.

Get early access to peptide careLow testosterone or missed periods can come from a failure at any link in the chain from brain to gonads. Hypothalamic amenorrhea shows the chain can go quiet from the top even when the ovaries are healthy.
So the first step is to ask which link has failed. Labs separate them: low LH and FSH with low testosterone points high in the chain, while high LH and FSH points to the gonads.8749

Kisspeptin-10 is the minimal, fully active fragment of kisspeptin, a hypothalamic hormone that triggers GnRH release and so starts the reproductive hormone chain. It is not FDA-approved.
In research it is used to study puberty, fertility and hypothalamic disorders, and as a possible trigger for egg maturation in IVF. People ask about it for testosterone and libido. It is not FDA-approved to treat any condition.
Studies show kisspeptin-10 raises LH, FSH and testosterone in healthy men. Clinical outcomes such as improved fertility or sexual function have not been shown for kisspeptin-10 itself.
It binds the KISS1R receptor on GnRH neurons, which release GnRH. GnRH makes the pituitary release LH and FSH, and those hormones drive the testes or ovaries.
Short-term studies report few adverse events, but they are small and brief. FDA notes immunogenicity and impurity concerns and limited safety information.
There is no FDA-approved dose. Research gave kisspeptin-10 into a vein or by pump under the skin, as single injections or timed infusions. With constant five-day exposure, LH and FSH fell back to placebo levels while testosterone stayed raised. Do not use an online chart to dose yourself; a clinician decides any use.
Kisspeptin acts upstream and makes the body release its own GnRH. Gonadorelin is GnRH itself and acts directly on the pituitary. Both are named on WADA’s list for males.
It is not FDA-approved. FDA lists it in category 2 for 503A compounding, with safety concerns, and it was not covered by the October 2026 enforcement discretion for six peptides. Products sold as research chemicals are not made or tested for people.
In males, yes. WADA’s 2026 Prohibited List names kisspeptin and its agonist analogues among testosterone-stimulating peptides, prohibited in and out of competition.
