Longer telomeres
A 2025 Biogerontology study found epitalon lengthened telomeres in normal human cells in a dish by switching on telomerase.4

Epithalon, AEDG (Ala-Glu-Asp-Gly)
Epitalon is a four-amino-acid peptide designed from a pineal gland extract, studied for 25 years for telomeres, melatonin and aging. Its most quoted human result belongs to a different molecule.

Epitalon, also spelled epithalon, is a four-amino-acid peptide (Ala-Glu-Asp-Gly) designed from epithalamin, an extract of the pineal gland. In cell studies it switches on telomerase and lengthens telomeres, and in older monkeys and people it restored the nightly rise in melatonin. It is studied for healthy aging. Most evidence comes from lab and animal work, largely from one research group.

Epitalon is studied for how it affects two things that change with age.
In cell studies it switched on an enzyme that rebuilds telomeres, the protective caps on chromosomes.47
It acts on melatonin production, the hormone that sets your sleep rhythm.39
Lab studies describe antioxidant, nerve-protective and anti-mutation effects.3
Epitalon’s research spans cells, animals and older adults, much of it from the group that developed it. No controlled human trials exist.
In normal human cells in a dish, it switched on telomerase and lengthened telomeres.47
Shows a mechanism
Old monkeys regained the evening melatonin rise, and mice given epitalon lived longer.910
Shows an effect in a body
Russian groups reported restored night melatonin in older adults with weak pineal function. The mortality study used the parent extract, not epitalon.116
Shows it works in people
A 2025 review in the International Journal of Molecular Sciences summarizes 25 years of research and calls its effects geroprotective. Most human studies come from one Russian group, so independent trials would add weight.3
The full guide to Epitalon
Every evening, a small gland near the center of your brain starts releasing melatonin, the hormone that tells the rest of your body the day is over. With age that signal fades. Older monkeys and older people make less melatonin at night, and the daily rhythm flattens. In St. Petersburg, a group led by Vladimir Khavinson spent decades asking whether the signal could be turned back up.
Their first tool was crude. Epithalamin is a peptide extract of bovine pineal glands, and in the group’s own studies it raised melatonin production in rats, improved immune measures, slowed tumors in several experimental models and restored reproductive function in old rats. Rats, mice and fruit flies given the extract lived longer.
Extracts are messy, so the team analyzed the amino acid makeup of epithalamin and built a short peptide from it: alanine, glutamic acid, aspartic acid and glycine, written AEDG. The same sequence was later found in the pineal gland’s own peptide mix. The developers designed one tetrapeptide for both the pineal gland and the retina, which they linked to the two organs sharing an embryonic origin.
Epitalon, epithalon and epithalone are all the same molecule. A 2025 review from Warsaw, written by a group outside the original program, describes 25 years of cell, animal and computer studies and calls its effects geroprotective. The same review notes how few studies have looked at the peptide’s basic chemistry, and that its known mechanisms may not be the only ones.1153
To follow the epitalon story you need one piece of cell biology. Telomeres are repeating caps of DNA at the ends of chromosomes, and they shorten a little each time a cell divides. When they get too short, the cell stops dividing. That ceiling is called the Hayflick limit, and telomerase is the enzyme that can rebuild the caps. Most adult cells keep it switched off.
In 2003, Khavinson’s group added epitalon to human fetal fibroblasts that had no telomerase activity. The cells began making the enzyme’s catalytic subunit, telomerase became active, and telomeres grew longer. A year later the group took fibroblasts that had stopped dividing at their 34th passage. With epitalon, their telomeres returned to early-passage length and the cells made 10 extra divisions, still going when the experiment ended.
The second thread runs through the pineal gland. In old female rhesus monkeys, epitalon stimulated evening melatonin production and normalized the daily cortisol rhythm, the stress hormone that should peak in the morning and fall by night. The 2025 review adds effects on interleukin-2, a T cell growth signal, on thymus cells and on several enzymes, including two that break down the nerve messenger acetylcholine.
Khavinson built a broader theory on this work. In his view, aging is partly a slow change in gene expression that reduces the body’s own regulatory peptides, and short peptides can nudge that expression back. It is a coherent idea, and it is still a theory. The independent review says plainly that it remains uncertain whether these are the only ways epitalon acts.78935
For about two decades, the telomere claim rested mostly on the developers’ own cell work. In 2025, a team at Brunel University London tested it again, with authors who declared no competing interests. They treated normal human epithelial cells and fibroblasts with epitalon, alongside two breast cancer cell lines, and measured telomere length, telomerase gene activity and enzyme activity.
In the normal cells, the result matched the Russian findings. Telomeres lengthened in a dose-dependent way, and the route was the expected one, with more hTERT, the gene for telomerase’s working part, and more telomerase activity. Independent replication matters in a field where one group produced most of the data, and this was a clean confirmation in a dish.
The cancer cells told a second story. Their telomeres also grew, and the authors traced that to ALT, the alternative lengthening of telomeres, a backup route some tumors use that does not depend on telomerase. Normal cells showed only a minor rise in ALT, so the authors read that response as specific to cancer cells.
Two things follow. The telomere effect is real enough to take seriously in cells. And a molecule that lengthens telomeres in cancer cells deserves care in anyone with a cancer history, even though mouse studies found fewer tumors, not more. A cell line is still a long way from a person, and none of the studies cited here measured telomere length in people taking epitalon.410
Melatonin is a timing signal more than a sleeping pill. It rises in the evening, peaks at night and falls by morning, and that curve helps set the clock for hormones and repair across the body. In both monkeys and people, aging lowers the night peak and the daily average and flattens the rhythm, a sign that the pineal gland is working less well.
The monkey work is the cleanest evidence here. In female rhesus monkeys of different ages, epitalon raised evening melatonin and brought the daily cortisol rhythm back toward a normal pattern in the old animals. A rhesus monkey runs its days much as we do, which makes these results more relevant than most rodent data.
In older people, the St. Petersburg and Kyiv groups reported that epithalamin and epitalon restored night melatonin in those whose pineal function was weak. In a related study, the pineal extract shifted the melatonin peak to around 3 a.m., the timing typical of young people, and a thymus hormone rose at night alongside it.
Keep the limits in view. These studies came from the developers’ network, their summaries do not give group sizes, and they report hormone levels rather than how well people slept or how they felt the next day. The authors described the preparations as having no side effects. None of the studies cited here is a controlled trial of sleep quality.11912
The most cited animal experiment ran for the life of the mice. Female mice received epitalon injections from six months of age until they died. Their weight, food intake, activity and behavior did not change, which matters because eating less alone can extend a mouse’s life.
What did change was the pace of aging. The treated mice kept their reproductive cycles longer, ran slightly cooler, showed slower free radical damage and lived longer. They also developed fewer spontaneous tumors, the opposite of what the telomerase worry would predict. The developers report longer lifespans in fruit flies too.
One of the more concrete results involves the eye. In rats with an inherited retinal degeneration, epitalon kept the retina working longer, and the developers report better visual function in patients with pigmentary retinal degeneration. That fits their idea that the same peptide speaks to the pineal gland and the retina, which develop from related tissue.
A longer-lived mouse is a reason to form a hypothesis about people. These studies came largely from labs tied to the program, and no large independent replication appears in the work cited here, so they explain the interest in epitalon without predicting what it would do for a person.105
The number you will see most often comes from a 2003 paper with a bold title: peptides of the pineal gland and thymus prolong human life. Researchers in St. Petersburg and Kyiv followed 266 older people for six to eight years. Groups given the pineal preparation had 1.6 to 1.8 times lower mortality than controls, and a group given pineal and thymus preparations every year for six years had 4.1 times lower mortality.
Look closer and one detail changes the picture. The pineal preparation was epithalamin, the extract, and the thymus preparation was thymalin, another extract. Epitalon, the four-amino-acid peptide sold today, was not what those people received. Online, the mortality figures travel attached to epitalon anyway.
The developers argue that epitalon reproduces the effects of epithalamin, which is a fair hypothesis for a peptide designed from the extract’s composition. But an extract holds many peptides, and a single tetrapeptide may or may not carry its full effect. None of the studies cited here repeats that long-term human follow-up with epitalon alone.
The study itself also needs context. The summary does not describe how people were assigned to groups, and the work came from the developers’ own institutes. Effects this large would need independent confirmation before anyone relied on them. So the most useful thing to remember about epitalon is that its strongest human evidence belongs to its parent extract.65
Set the extract aside, and the human record for epitalon itself is short. There are the melatonin studies in older adults, where epitalon restored night-time melatonin in people with weak pineal function. And there are the developers’ reports of better visual function in patients with retinal degeneration.
Everything else, from telomeres to lifespan, comes from cells and animals. That is not unusual for a peptide, but it means the claims you read about reversing biological age or adding years have no controlled human trial behind them. The 2025 review is upbeat about the science and still describes epitalon, in its title, as a peptide with promising properties.
Two gaps stand out. Independent groups have confirmed parts of the cell biology, but not the human results. And none of the studies cited here followed people on epitalon with modern endpoints such as measured telomere length, validated sleep scores or hard health outcomes. Those are the experiments that would turn an interesting peptide into a known one.11534
The reported safety record is quiet. The Russian clinical studies did not describe serious side effects, and the authors of the melatonin work described the pineal preparations as having none. In lifelong mouse studies, epitalon did not change weight, eating or behavior, and spontaneous tumors went down rather than up.
The open question is theoretical but serious. Many cancers keep growing by keeping telomerase switched on, so any compound that switches it on deserves caution. The 2025 Brunel finding that cancer cells lengthened their telomeres through a different route on epitalon adds a specific reason for care in anyone with a cancer history.
Quality is the other risk. FDA’s April 2026 compounding page noted limited human safety data and the possibility of an immune reaction to impurities in the peptide. A prescription filled by a licensed pharmacy is a different product from an unlabeled vial. People who are pregnant or breastfeeding, or living with kidney or liver disease, should also talk with a clinician first.51110415
No blood test measures epitalon itself, and none of these markers is a validated readout of biological age. What a panel gives you is general health context a clinician can review before and after. Fasting glucose is a simple marker of metabolic health, and the body clock that epitalon is studied for helps regulate it. A baseline and a retest show whether your metabolism shifted over the same period.
ALT is a liver enzyme that leaks into the blood when liver cells are under strain, and eGFR estimates how well your kidneys filter. Neither is a known target of epitalon, and no epitalon-specific monitoring protocol exists in the studies cited here, but both are common baseline checks a clinician may want before a new prescription. hs-CRP tracks low-grade inflammation, one of the hallmarks of aging that no single peptide can fix alone.
This is how Vitals Vault approaches peptide care. A blood panel comes first, a licensed clinician reviews your results and history and decides whether epitalon fits, and the course ends with a retest of the same markers. The clinician sets the form and length of the course.139
On October 29, 2026, NCPA reported that FDA granted enforcement discretion allowing 503A compounding pharmacies to prepare epitalon, and five other peptides, for individual patients with a prescription. There is no end date, and final rulemaking is pending.
Epitalon is still not an FDA-approved drug, which means no company has put it through the trials an approval requires. FDA’s April 2026 safety page noted limited human data, and vials sold online as research chemicals are not covered by the new policy.141516
Epitalon is a tiny peptide with a long, unusual history. In normal human cells it switched on telomerase in both developer and independent studies, in old monkeys and older adults it restored the nightly melatonin rise, and in mice it extended life. Its most famous human result came from the extract it was designed to copy, and the cancer-cell findings argue for care.
If you are considering it, start with the parts of aging you can measure and move: sleep timing, blood sugar, inflammation and the basics of exercise and light. Then the question worth asking a clinician is specific. Which of my aging markers would epitalon plausibly change, and how would we know if it did?411106
A responsible start is a health history, a baseline blood panel and a clinician’s decision on whether epitalon fits your goals and your cancer risk. The older studies gave epitalon as a course rather than continuously, and the prescriber sets the form and length.
On timing, the studies offer only partial guidance. The melatonin changes in older adults and monkeys were measured over a course of treatment, while the telomere and lifespan effects come from cells and lifelong mouse studies that do not translate into a human timeline. Judge it by markers you can retest, and by whether your sleep rhythm changes.119

The human mortality study behind that claim gave older adults epithalamin, the pineal extract, and thymalin, a thymus extract. None of the studies cited here measured lifespan in people taking epitalon itself. In mice, it did extend life.610
Longer telomeres let cells keep dividing, which is also what cancer cells need. In a 2025 study, epitalon lengthened telomeres in breast cancer cell lines through a telomerase-independent route, so a cancer history calls for a clinician’s view first.4
It contains no melatonin. In studies it acted on the pineal gland so that the body made more of its own melatonin in the evening, restoring a rhythm rather than adding a fixed dose of hormone.911
Most of it does come from one Russian group, which is a real limitation. But an independent team in London confirmed in 2025 that epitalon lengthens telomeres in normal human cells, and a Warsaw group reviewed the field.43
Epitalon is prescribed as an injection under the skin or a nasal spray. The older Russian studies gave it as a course of treatment rather than continuously. Your prescriber sets the form, dose and schedule.
There is no FDA-approved epitalon dose, and the human dosing data are thin. FDA’s 2026 review found three human studies: two gave epitalon as a spray under the tongue to women working night shifts, and one gave injections beside the eye in people with congenital retinitis pigmentosa. The table shows the sublingual regimen, a small daily amount for 20 days. FDA found no clinical study of the injection under the skin that compounders propose. Higher injected amounts quoted online come from clinic websites, not trials. Your clinician sets your dose and the length of any course.12
The table shows the one human sublingual regimen: a spray under the tongue, twice a day for 20 days, in women aged 40 to 50 working night shifts with low melatonin output. A separate human study gave injections beside the eye for congenital retinitis pigmentosa, which says nothing about injection under the skin. No FDA label or trial in people sets a dose for the injection form that compounding pharmacies prepare. Your clinician sets the actual dose.

Vitals Vault peptide care starts with a blood panel and a licensed clinician, and ends with a retest.
Get early access to peptide careAging is not one process. Researchers describe a set of hallmarks that drive it, including telomere shortening, mitochondrial decline, chronic inflammation and disrupted signaling. They feed into each other, which is why no single molecule can address aging on its own.
The measurable parts of aging come first: sleep timing, blood sugar, inflammation and the basics of exercise and light. A clinician can ask which of your aging markers a peptide would plausibly change, and how you would know.133
A blood test before you start and a retest after shows what actually changed for you. It is how Vitals Vault approaches every treatment.

Epitalon is studied for healthy aging: telomere length, melatonin and sleep rhythm, and cell protection. It is not FDA-approved to treat any condition.
Research reports longer telomeres in human cells, a restored nightly melatonin rise in old monkeys and older adults, longer life in mice, and antioxidant and nerve-protective effects. Controlled human trials are still missing.
In cell studies it switches on telomerase, the enzyme that rebuilds the protective caps on chromosomes. It also acts on the pineal gland’s melatonin production and on gene activity, and it may work through other routes not yet mapped.
In human cells in a dish, yes. Studies from 2003 and 2025 found longer telomeres through telomerase. None of the studies cited here tested whether the same happens in a person.
Both. They are two spellings of the same peptide, Ala-Glu-Asp-Gly, and you will also see epithalone.
Epithalamin is a peptide extract of the pineal gland. Epitalon is a single four-amino-acid peptide designed from that extract’s composition. The long-term human mortality study used epithalamin, not epitalon.
The Russian studies did not report serious side effects, but modern safety trials have not been published. Because it switches on telomerase, people with a history of cancer should talk to a clinician first.
There is no FDA-approved dose. One human trial used a small daily spray under the tongue for 20 days, and a separate study tested injections beside the eye for retinitis pigmentosa. FDA found no clinical study of injection under the skin, so neither sets a dose for that form. Your clinician sets your dose and schedule after a review of your history and labs.
No. Epitalon is not FDA-approved for any use. Since October 29, 2026, FDA allows 503A compounding pharmacies to prepare it for individual patients with a prescription, under enforcement discretion, while final rulemaking is pending.
A licensed prescriber can now order compounded epitalon from a 503A pharmacy for a specific patient. Vials sold online as "research use only" are not made for people and are not covered by FDA’s October 2026 policy.
