## The answer, in short

The peptides most studied for longevity are epitalon, which switches on telomerase in cells and restored night melatonin in older adults, the mitochondrial peptides humanin and MOTS-c, which track healthy aging and respond to exercise, and glutathione, the cell’s own antioxidant. None has a controlled human trial showing longer life, and growth hormone-raising peptides may pull the other way.

## What would it take for a peptide to slow aging?

Somewhere in their fifties, a lot of people start reading about telomeres. Recovery takes longer, sleep gets thinner, the waistline creeps, and a friend mentions a peptide that is said to lengthen the caps on your chromosomes. It is a reasonable thing to hope for, and a reasonable thing to check.

Aging is not one process, which is the first problem for any single molecule. The 2023 update of the hallmarks of aging, the framework most researchers use, lists twelve of them, from telomere attrition and mitochondrial dysfunction to chronic inflammation and deregulated nutrient sensing. Each hallmark has to meet three tests, appearing with age, speeding aging when pushed, and slowing aging when corrected.

Seen that way, a longevity peptide is a bet on one or two hallmarks. Epitalon is a bet on telomeres and the body clock. Humanin and MOTS-c are bets on the mitochondria. Glutathione is a bet on oxidative stress. The honest question for each is how far the evidence has traveled from a dish or a mouse toward a person living longer and better.[1](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-lopezotin-2023)

## Where longevity peptides act

The peptides in this chapter sit at four points in the aging network. Most of them are copies or fragments of molecules the body already makes, and several fall with age, which is much of their appeal.

That appeal has a catch. A molecule that falls with age might be a cause of aging, a consequence of it, or a bystander. Topping it back up only helps in the first case, and the way to tell is a trial in people that measures outcomes, not just levels. Keep that test in mind through every section below.[1](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-lopezotin-2023)

### Redox balance

Glutathione is the main antioxidant cells make for themselves, and older adults make it more slowly.[17](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-wu-2004)[18](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-sekhar-2011)

## Longevity peptides compared

The table lines up the longevity peptides by mechanism and by the best evidence behind each. Notice how often the strongest human finding measures a natural level, a gene variant or a hormone, rather than an outcome in people given the peptide.

Peptides studied for longevity
| Peptide | How it works | Best evidence | Evidence level |
| --- | --- | --- | --- |
| Epitalon | Switches on telomerase; supports night melatonin | Human cell replication; small melatonin studies in older adults | Early human data |
| Humanin | Blocks the cell-death protein Bax; improves insulin action via the brain | Higher levels in children of centenarians; lifespan in worms | Early research |
| MOTS-c | Switches on AMPK in muscle; rises with exercise | Exercise raises it in people; late-life benefit in old mice | Early research |
| Glutathione | Buffers oxidative stress inside cells | Six-month randomized trial raised body stores | Human trials |

## Epitalon and the extract it was built from

Epitalon is four amino acids, alanine, glutamic acid, aspartic acid and glycine, designed in St. Petersburg from the makeup of epithalamin, an extract of bovine pineal glands. In 2003 its developers showed it switched on telomerase in human cells that had none. In 2025 an independent team at Brunel University London repeated the experiment and saw the same dose-dependent telomere lengthening in normal cells.

The same Brunel study added a reason for care. Cancer cells lengthened their telomeres too, mostly through a backup route called ALT. Lifelong mouse studies, for their part, found fewer spontaneous tumors and longer lives on epitalon, and in older people with weak pineal function the developers reported that epitalon restored night melatonin.

The number you will see most often belongs to the extract. A 2003 study followed 266 older people for six to eight years and reported 1.6 to 1.8 times lower mortality in groups given the pineal preparation. That preparation was epithalamin, the extract, and nobody has run the same long-term study with epitalon itself.

The melatonin work is the most human part of the epitalon story, and it is small. The studies came from the developers’ network, measured hormone levels rather than how people slept, and described no side effects. Whether epitalon helps anyone sleep better has not been tested in a controlled trial.[2](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-khavinson-2003-telomerase)[3](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-aldulaimi-2025)[5](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-anisimov-2001)[6](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-korkushko-2007)[7](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-khavinson-2003)

## Humanin and MOTS-c, messages from your mitochondria

For decades the small circle of DNA inside each mitochondrion was thought to code only for parts of the energy machinery. Then humanin, found in 2001 in a search for what keeps neurons alive, turned out to be coded there. Inside the cell it binds Bax, a protein that punches holes in mitochondria to start cell death, and holds it back.

Humanin falls with age in mice and people. In 2020 a large collaboration found that it extended lifespan in worms, that a potent analog improved metabolic health in middle-aged mice, and that children of centenarians had much higher circulating humanin than people without that family history. That last finding is an association, and it cannot say whether humanin causes longer life or marks it.

MOTS-c, found in 2015, acts mainly on muscle, switching on AMPK, the cell’s fuel gauge. Exercise raises it in people, in muscle and in blood, and in mice it improved physical capacity even when started at an age equivalent to late old age. Neither peptide has a published human treatment trial. The most reliable known way to raise both is already in your shoes, since resistance training raised humanin in muscle by 35% in men with impaired glucose control.[8](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-hashimoto-2001)[9](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-guo-2003)[11](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-yen-2020)[13](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-lee-2015)[14](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-reynolds-2021)[12](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-gidlund-2016)

## Glutathione and a factory short on parts

Glutathione is three amino acids, glutamate, cysteine and glycine, and the most abundant antioxidant cells make for themselves. Its link to aging is one of the best measured in this chapter. In 2011 a Baylor team traced its production in older and younger adults and found the older group had about half the red cell glutathione and made it at roughly a third of the rate.

The cause was supply. After two weeks of cysteine and glycine, the building blocks, the older adults’ glutathione rose by 94.6% and oxidative stress fell to the level of the young group. A later 16-week randomized trial of glycine plus NAC in 24 older adults improved glutathione, inflammation, insulin resistance, gait speed and strength.

Glutathione itself also has trial data. In a six-month randomized trial, oral glutathione raised levels in blood and immune cells, and they fell back to baseline within a month of stopping. What no trial has shown is that extra glutathione helps people who were not low to begin with.[17](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-wu-2004)[18](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-sekhar-2011)[19](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-kumar-2023)[20](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-richie-2015)

## The growth hormone trade-off

Many of the most popular anti-aging peptides, sermorelin, ipamorelin, CJC-1295, MK-677, are designed to raise growth hormone and IGF-1, because both fall with age and youth feels like more of them. Yet across model organisms, turning the growth pathway down tends to lengthen life, which is why deregulated nutrient sensing sits on the list of hallmarks.

Humanin makes the trade-off visible. In 2014, researchers found that Ames dwarf mice, which lack growth hormone and live unusually long, had high humanin, while mice engineered to make extra growth hormone had low humanin and shorter lives. Giving growth hormone or IGF-1 lowered humanin in mice, and it did the same in human subjects.

People offer a natural experiment too. In an Ecuadorian community with inherited growth hormone receptor deficiency, followed for 22 years, affected people had one nonlethal cancer and no diabetes, against 17% cancer and 5% diabetes among their relatives. Their insulin sensitivity was markedly higher.

None of this means growth hormone peptides shorten life, and nobody has measured humanin in people taking them. It does mean that longevity and growth are not the same goal, and a protocol built on one may cost something on the other. That is a question worth asking before anyone stacks a growth peptide with a longevity peptide and expects them to add up.[1](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-lopezotin-2023)[10](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-lee-2014)[16](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-guevara-2011)

## What else moves the same system, and the labs to check

The striking thing about the mitochondrial peptides is how much of their story runs through exercise. MOTS-c rises when you move, humanin rises in trained muscle, and a variant of the MOTS-c gene raised diabetes risk mainly in the least active men in a Japanese cohort. Training is the oldest longevity intervention there is, and these peptides look like part of how it works.

Sleep timing matters for the same reason epitalon was studied. Melatonin is a timing signal, and aging flattens its nightly rise. Protein matters for glutathione, because cysteine and glycine set the pace of production.

Labs turn hallmarks into numbers you can retest. HbA1c and fasting insulin track the nutrient-sensing system that growth hormone and the mitochondrial peptides both act on. hs-CRP measures the low-grade inflammation of aging. GGT reflects the oxidative load that spends glutathione.

None of these levers needs a prescription, and each has more human evidence for healthy aging than any peptide in this chapter. A peptide, if a clinician thinks one fits, belongs on top of that foundation, with a baseline panel before the course and a retest of the same markers after it.[14](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-reynolds-2021)[12](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-gidlund-2016)[15](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-zempo-2021)[6](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-korkushko-2007)[17](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-wu-2004)[21](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-whitfield-2001)

## Where longevity peptides stand in 2026

On October 29, 2026, NCPA reported that FDA granted enforcement discretion allowing 503A compounding pharmacies to prepare epitalon and MOTS-c, among six peptides, for individual patients with a prescription. Neither is FDA-approved, and final rulemaking is pending. FDA’s April 2026 safety page had noted limited human data for these substances. Humanin is not covered and has no regulated prescription route. Glutathione is sold as a supplement by mouth, and pharmacies compound injections with a prescription under FDA’s 503A framework.[22](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-ncpa)[23](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-fda-bulks)[24](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-fda-503a)

## What this means for you

The longevity peptides have real biology behind them, telomerase in a dish, mitochondrial signals that track long-lived families, an antioxidant older bodies make too slowly. What none of them has is a human trial showing that people given the peptide live longer or age better, and the most quoted mortality figure belongs to an extract.

The best place to start is the part you can measure and move, which is training, sleep timing, protein and the markers that track them. Then the question for a clinician is sharp. Which hallmark of aging is this peptide supposed to change in me, which lab would show it, and does anything else I take push in the opposite direction?[7](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-khavinson-2003)[11](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-yen-2020)[18](https://www.vitalsvault.com/peptides/peptides-for-longevity#source-sekhar-2011)

## Questions

What is the best peptide for longevity?

No peptide has been shown to extend human life. Epitalon has the longest research history and independent cell data on telomeres. Humanin and MOTS-c have strong animal data and human links to healthy aging. Glutathione has the most human trial data, mostly on raising body stores.

Does epitalon really lengthen telomeres?

In human cells, yes. The developers reported it in 2003 and an independent team confirmed it in 2025. The same 2025 study found cancer cells also lengthened their telomeres. No controlled human trial has measured telomere length on epitalon.

What are mitochondrial peptides?

Short peptides coded in the small genome inside mitochondria. Humanin, found in 2001, protects cells from self-destruction. MOTS-c, found in 2015, acts on muscle metabolism and rises with exercise. Both tend to fall with age.

Do growth hormone peptides help longevity?

Not necessarily. In mice, lower growth hormone signaling goes with longer life, and giving growth hormone or IGF-1 lowered humanin, a peptide linked to longevity, in mice and people. Raising growth hormone and slowing aging are different goals.

Is glutathione an anti-aging peptide?

It is a three-amino-acid peptide every cell makes, and older adults make it more slowly. Building blocks restored low levels in older adults, and oral glutathione raised body stores in a six-month trial. Benefit in people who are not low is unproven.

Can exercise raise my own longevity peptides?

Yes, for the mitochondrial ones. Exercise raised MOTS-c in human muscle and blood, and twelve weeks of resistance training raised humanin in muscle by 35% in men with impaired glucose control.

What labs should I track for longevity?

HbA1c and fasting insulin show how your metabolism is aging. hs-CRP measures low-grade inflammation. GGT reflects oxidative load on glutathione. A baseline and a retest of the same markers are the clearest way to see whether anything changed.
